三种分离皮肤方法提取表皮下大疱病相关抗原比较

Comparison of three techniques of separating skinfor extracting antigens related to S A B D

目的 比较提取表皮下大疱病相关抗原的三种分离皮肤方法。方法 采用盐裂法、 Ｅ Ｄ Ｔ Ａ 分离法、热分离法提取真、表皮抗原，免疫印迹检测提取物中抗原的活性，结果 三种方法均可用于 Ｓ Ａ Ｂ Ｄ 抗原的提取，但热分离法在所提取抗原的敏感性上明显优于其他两种方法。结论 热分离法对抗原活性影响最小，值得优先选择和运用。

Objective To co mpare three methods extracting antigens related to S A B D from hu man skin . Methods Three techniques of 1 m ol· L- 1 Na Cl, 0 1 mol· L- 1 E D T A and heat separated skin were used toisolate dermal and epidermal antigens . The activity of antigens from extract was detected by im munoblotting . Results Three techniques might extract S A B D antigens , but the sensitivity of antigens from heat separatedskin was higher than the other two . Conclusion Heat seperation technique produces the least influence onantigen activity and is the first choice .
