摘要
[目的]利用环介导等温扩增技术快速检测志贺氏菌。[方法]以志贺氏菌侵袭性质粒抗原H基因(ipaH)作为靶序列,设计引物,优化Mg2+浓度、Bst酶浓度等反应条件,建立LAMP反应体系。[结果]确定了环介导等温扩增技术检测志贺氏菌的适宜反应条件;该法检测志贺氏菌的灵敏度为62 cfu/ml。[结论]该研究初步建立了一种利用LAMP快速检测志贺氏菌的方法,为食品中志贺氏菌快速检测构建了一个技术平台。
[Objective] The aim was to detect Shigella by using the loop-mediated isothermal amplification(LAMP) technology.[Method] The sequences of ipaH of Shigella was used as target sequences to design primers,the reaction conditions such as Mg2+ concentration,Bst DNA polymerase concentration were optimized for establishing LAMP system.[Result] The reaction conditions were optimized and the sensitivity of LAMP assay was 62 cfu/ml.[Conclusion] LAMP technology for detection of Shigella was developed,and a technology platform was constructed for the rapid detection of shigella in food.
