摘要
以引种的黑木相思优株为材料,采用正交设计进行组织培养试验。研究结果表明:最适宜的黑木相思外植体诱导培养基为改良MS+BA1-5~2-3mg·L-1+NAA0-2~0-4mg·L-1,诱导启动率达90%,继代增殖培养基以改良MS+BA2-0~3-0mg·L-1+NAA0-2~0-3mg·L-1最佳,增殖率达7倍,生根诱导的培养基为改良MS+NAA0-5mg·L-1+IBA0-2mg·L-1,生根率达90%,试管苗移植成活率达80%。
The study results reveal that the most suitable induction culture medium of Acacia melanoxylon explants is the improved MS+BA1 5~2 3mg·l -1 +NAA0 2~0 4mg·l -1 ,the induction starting rate reaches 90%,the optimum culture medium for the generation continuing multiplication is the improved MS+BA2 0~3 0mg·l -1 +NAA0 2~0 3mg·l -1 ,the multiplication rate reaches 7times,the rooting induction culture medium is the improved MS+NAA0 5mg·l -1 +IBA0 2mg·l -1 ,the rooting rate reaches 90% and the test tube plant transplanting survival rate reaches 80%.
