摘要
目的左金丸方药仿生提取工艺的探索、改进与优化。方法以不同提取溶剂提取左金丸方药,并通过酸种类和pH值变化对工艺进行优化,采用高效液相色谱法对左金丸主要生物碱提取率进行测定和镇痛模型的药效学实验加以验证。结果用仿生提取法优化工艺后,相同时间内,人工胃液提取率最强(人工胃液>60%乙醇提液>人工肠液>水提液),且较之其他方法可提高近2倍或2倍以上,与水提和醇提组相比有显著性差异(P<0.05,P<0.01),且在提取的较短时间内可达到峰值。优化实验证明仿生提取液中采用盐酸调节pH值效果最好。镇痛实验表明胃液左金组与正常对照组相比,痛阈提高率存在显著性差异(P<0.05)。结论用仿生提取液作为提取工艺稳定可行,可作为左金丸提取工艺并为日后的剂型改造奠定基础。
AIM To research the bionics extraction technology of Zuojin Pill and its improvement and optimization.METHODS To extract Zuojin Pill with different extracting agents and optimize it through acid variety and the change of pH value.Using the method of HPLC to determine the extracting percentage of main alkaloid of Zuojin Pill and verify experiment of pharmacodynamics of analgesia model.RESULTS After using the extract method of bionics for optimization,within the same time,extracting percentage of simulated gastric fluid was the best(artificial gastric juice 60%ethanol extracting solution artificial intestinal juice water extracting solution),and that could increase twice or more than twice than other methods and had significant difference compared with water and 60%ethanol extracting solution group(P0.05,P0.01),at the same time,it could come to peak value quickly within the shortest time.The results of the verification demonstrated that,in the biomimetic extracting solution,the effect was the best by adopting hydrochloric acid to set the value of pH.The results of the analgesia indicated that,artificial gastric juice group compared with the regular group,the increase rate of pain threshold existed significant difference(P0.05).CONCLUSION It is stable and feasible that using bionics extracting solution as extraction process.It can be used as Zuojin Pill extraction process and settle the base for dosage form alteration in the future.
