摘要
目的:研究环磷酰胺的免疫抑制作用机制。方法:以大鼠胸腺为对象,采用细胞凋亡的原位特异性染色及DNA凝胶电泳的方法,观察不同剂量、不同时间环磷酰胺处理后胸腺细胞的凋亡现象。结果:剂量为20和70mg/kg的环磷酰胺可引起广泛的胸腺细胞凋亡。该作用最早发生在环磷酰胺处理后的8h,12~24h最明显,48h消失。结论:环磷酰胺的免疫抑制作用可能是通过诱导免疫细胞凋亡而产生的。
Objective:To understand the mechanism of immunosuppression of cyclophosphamide. Methods: Investigate if cyclophosphamide produces the immunosuppression by the induction of apoptotic cell death in thymus .Results: 20 and 70 mg/kg cyclophosphamide could induce markedly apoptosis in rat thymus at 12 h after treatment ,the apoptotic cell death took place at 8~24 h and disappeared at 48 h after cyclophosphamide treatment. Conclusion: Cyclophosphamide may causes immunosuppression through the induction of apoposis of thymocyte.
