摘要 目的:检测肿瘤细胞条件培养液中VEGF的表达量。方法:在表达及分离纯化VEGF的基础上,成功地制备了VEGF抗体,以夹心法测定VEGF。结果:VEGF测定范围为002~20000ng/ml,灵敏度可达002ng/ml,批内及批间变异均小于10%,回收率平均为1024%。其中人胃癌细胞MGC803、BGC823、乳癌MCF7及肝癌BEL7402分泌上清中VEGF的含量分别为650、045、090及145ng/ml。结论:不同的肿瘤细胞系其VEGF的分泌量是不同的,应用此方法可以检测肿瘤病人血清中的VEGF。 Objective: To detect expression level of VEGF is important for determining biological behaviour of tumor cells. Methods: Based on isolation, purification and expression of VEGF,successfully prepared anti VEGF antibody and developed a sandwich radioimmunoassay for VEGF. Results: Detectable range of VEGF is between 0.02 and 200.00 ng/ml.The sensitivty or minimal detectable concerntration dose was difference of sample in the same group and among groups were all less than 10 percent. By the assay it is indicated that all four kinds of tumor cell lines secreted VEGF, MGC803, BGC823, MCF 7 and BEL 7402 separeately secreted 6.50,0.45, 0.90 ,1.45 ng/ml. Conclusion: VEGF can be detected in the serum of tumor patient with this assay.
