摘要
目的:观察转化生长因子B1(Transforming gowth factor-β11TGF-β1)在阿霉素肾病(Doxorubicin.induced nephropathy DIN)大鼠疾病早期的表达及辛伐他汀对TGF-β1表达的影响。方法:将100只6周龄雄性sD大鼠随机分为正常组、肾病组和辛伐他汀治疗组3组,第7、21、42天检测各组大鼠胆固醇、甘油三酯,24小时尿蛋白定量、尿TGF-β1以及肾组织中TGF-β1。结果:(1)、21天辛伐他汀组与肾病组比较各项检查均无显著差异(P>0.05);第42天辛伐他汀组尿蛋白低于肾病组(P<0.05),有显著差异;尿TGF-β1、肾组织TGF-β1、胆固醇、甘油三酯有非常显著差异(P<0.01);(2)、肾病组肾组织TGF-β1与尿蛋白、尿TGF-β1成正相关。结论:TGF-β1与MCNS尿蛋白的发生有关;辛伐他汀可降低血脂及尿蛋白,降低TGF-β1的表达,减轻肾脏损害,延缓疾病进展。
Objective:To observe the expression of transforming growth factor-β1(TGF-β1)in early stage of Doxorubicin.induced nephropathy(DIN)rats and Effects of Simvastatin on the expression of TGF-β1.Methods:100 6-week-old male SD rats were randomly divided into three groups:normal,nephrotic group and simvastatin group.Cholesterol、triglycerides、the 24-hour urine protein counts、urinary TGF-β1 and renal tissue TGF-β1 of all rats were measured on 7,21、42 days after injection.Results:(1)21 days of Simvastatin group and the nephrosis group were not significantly different(P0.05);42 days of urinary protein were significantly different than kidney simvastatin group(P0.05);In urinary TGF-β 1,renal TGF-β 1,cholesterol,triglycerides have significant difference(P0.01);(2)nephrosis group TGF-β1 in renal tissue and urine protein and urine TGF-β1 were positive correlation.Conclusion:TGF-β1 was related with MCNS urine protein occurrencel-simvastatin can reduce blood lipids and urinary protein,reducing the expression of TGF-β1,to reduce kidney damage,delay disease progression.
