摘要
以崇明岛53只崇明白山羊、30只徐淮山羊、37只杂山羊为实验材料,利用直接测序寻找心脏型脂肪酸结合蛋白基因(H-FABP)部分exon2及intron2序列上的SNPs,PCR-RFLP分析其多态性。结果表明:存在3个SNP位点(A1017G、C1115T、G999-)。A1017G通过HaeⅡ-RFLP得到BB、Bb、bb 3种基因型,除徐淮山羊外,崇明白山羊和杂山羊处于Hardy-Weinberg非平衡状态,3个群体均为低度多态(PIC<0.25)。C1115T未引起酶切位点改变。G999-通过SauⅠ-RFLP得到DD、Dd、dd 3种基因型,崇明白山羊处于Hardy-Weinberg非平衡状态,为低度多态(PIC<0.25);徐淮山羊和杂山羊只有DD基因型,呈严重的偏态,没有多态性。本研究表明H-FABP在不同山羊品种中具有单核苷酸多态性,可进一步作为候选基因分析其与肌内脂肪含量性状的关联性。
Direct sequencing was used to find the SNPs at the H-FABP gene in 53 Chongming white goats,30 Xuhuai goats and 37 hybridized goats from Chongming Island.PCR-RFLPs were established to differentiate the SNPs.The results showed that there were three SNPs(A1017G,C1115T,G999-).A1017G can be analysised by HaeⅡ-RFLP,and there were BB,Bb,bb three genotypes,which suggested that the allele frequencies and genotype frequencies of H-FABP gene were not in Hardy-Weinberg equilibrium in the Chongming white goats and the hybridized goats except the Xuhuai goats.The PIC values were all low(PIC0.25).C1115T didn't change the restriction enzyme cutting sites.G999-can be analysised by SauⅠ-RFLP,and there were DD,Dd,dd three genotypes,which suggested that the allele frequencies and genotype frequencies of H-FABP gene were not in Hardy-Weinberg equilibrium in the Chongming white goats,the PIC value was low(PIC0.25);There was only DD genotype in the Xuhuai goats and the hybridized goats,the skewness is serious and there were no polymorphisms.It meant that the fragment of H-FABP had polymorphisms,which could be used as a candidate gene associated gene with phenotypic traits like intramuscular fat content in different goat populations.
