维生素E对硫丹染毒小鼠脾细胞因子基因转录的影响 被引量:1

摘要 目的为了探讨维生素E对硫丹染毒ICR小鼠脾细胞因子白细胞介素-2(IL-2)、白细胞介素-6(IL-6)和γ-干扰素(IFN-γ)基因转录的影响,我们设计了该实验。方法将24只ICR小鼠随机分成对照组、硫丹染毒组(染毒组)和染毒同时给予维生素E(VE)组(实验组),所用硫丹和维生素E的剂量分别是:0 mg/(kg.d)和0 mg/(kg.d)(对照组)、0.8 mg/(kg.d)和0 mg/(kg.d)(染毒组)和0.8 mg/(kg.d)和100 mg/(kg.d)(实验组)。灌胃21 d后,取其脾,用Real-Time PCR法检测其脾脏IL-2、IL-6、IFN-γ基因转录的变化。结果染毒组小鼠脾细胞因子IL-2和IL-6 mRNA水平与对照组相比明显升高,IFN-γmRNA水平虽有升高趋势,但无显著变化。实验组小鼠脾IL-2和IL-6 mRNA水平明显低于染毒组,IFN-γmRNA水平虽有降低趋势,但与染毒组相比无显著变化。结论上述结果说明,硫丹能促进小鼠脾脏细胞因子IL-2和IL-6的基因转录,但对IFN-γ基因转录没有显著影响;维生素E能缓解或部分缓解硫丹的作用。 Objective To investigate the effects of vitamin E on the transcription of spleen IL-2,IL-6 and IFN-γ genes in mice treated with endosulfan. The treatment was assigned 3 groups,and the level of both endosulfan and vitamin E were 0 mg/ ( kg·d) and 0 mg/ ( kg· d) ( control group), 0. 8 mg/ ( ks·d) and 0 mg/ ( kg· d) ( endosulfan-treatment group) ,0.8 mg/ ( kg·d) and 100 mg/ ( kg" d) ( experimental group) , respectively. Spleens were collected to be analyzed the mRNA of IL-2,IL-6 and IFN-γ genes by Real-Time PCR method. The results showed that the mRNA levels of IL-2 and IL-6 gene in the endosulfan-treatment group were significantly higher than that of control group; although IFN-γ mRNA level in endosulfan-treatment group had an increasing tendency compared with control group,there was no significant difference between those two groups. The mRNA levels of IL-2 and IL-6, in the experimental group were significantly lower than that of endosulfan-treatment group,while the mRNA level of IFN-γ gene had a decreasing tendency in experimental group compared with endosulfan-treatment group,but there was no significant difference between those two groups. The results indicated that endosulfan can induce the gene transcriptions of spleen IL-2 and IL-6, but there is no significant effect on the gene transcription of spleen IFN-γ in mice, while vitamin E can alleviate or partly alleviate the endosulfan actions.
