高糖、胰岛素下调心肌细胞脂联素表达

摘要 目的观察心肌细胞能否表达脂联素(APN),并探讨高糖、高胰岛素状态下心肌细胞APN表达的变化。方法 Sprague-Dawley(SD)大鼠的乳鼠心肌细胞原代培养,分为葡萄糖6组:高渗组、5.5 mmol/L组、20 mmol/L组、25 mmol/L组、30 mmol/L组及50 mmol/L组。胰岛素6组:0 mol/L组、10-10 mol/L组、10-9mol/L组、10-8 mol/L组、10-7 mol/L组及10-6 mol/L组。干预48h后,用RT-PCR法测定心肌细胞APN-mRNA表达的差异,并检测心肌细胞APN蛋白表达的变化。结果在葡萄糖浓度小于30 mmol/L组中,心肌细胞APN表达随葡萄糖浓度的增加而减低(P<0.05),在大于30 mmol/L组中,APN表达无显著差异;渗透压对于心肌细胞APN表达无影响;在胰岛素浓度低于10-7 mol/L组中,心肌细胞APN表达随胰岛素浓度的增加而减低(P<0.05),在高于10-7 mol/L组中,心肌APN表达无显著差异。结论心肌细胞能够表达脂联素。在一定范围内,随葡萄糖、胰岛素浓度增加,心肌细胞APN的表达水平下降。 【Objective】 To study the influence of high concentration of glucose or insulin on the expression of adiponectin in cardiomyocytes.【Methods】 Primary culture the myocardial cells from the hearts of 2~3 days' neonatal Sprague-Dawley(SD) rats.There are 6 groups in glucose intervention part,including hypertonic control group,5.5 mmol/L glucose group,20 mmol/L glucose group,25 mmol/L glucose group,30 mmol/L glucose group,50 mmol/L glucose group.There are 6 groups in insulin intervention part,including 0 mol/L insulin group,10-10 mol/L insulin group,10-9 mol/L insulin group,10-8 mol/L insulin group,10-7 mol/L insulin group,10-6 mol/L insulin group.After 48 hours of incubation,we examined the expression of adiponectin by RT-PCR and western blot.【Results】 The expression of adiponectin decreased with increase of glucose concentration in concentration lower than 30 mmol/L(P 0.05).The osmotic pressure can not influence the adiponectin expression in cardiomyocytes.The expression of adiponectin decreased with increase of insulin concentration in concentration lower than 10-7 mmol/L(P 0.05).【Conclusion】 The cardiomyocytes can express adiponectin.In some concentration range,the expression of adiponectin decreased with increase of glucose and insulin concentration.
