摘要
目的探讨醛固酮对H9C2细胞胶原表达的影响。方法以培养的H9C2细胞为模型,观察100 nM醛固酮刺激H9C2细胞后0、1、3、6、12、24及48 h后盐皮质激素受体、转化生长因子β、结缔组织生长因子、Ⅰ型和Ⅲ型胶原的表达情况。结果免疫印记检测可见醛固酮刺激H9C2细胞3 h开始,盐皮质激素受体表达增高,6 h达高峰。Real-time PCR检测结果显示,醛固酮刺激后,转化生长因子β和结缔组织生长因子的表达在6 h开始增高,12 h达高峰。Ⅰ型和Ⅲ型胶原的表达在6 h开始增高,24 h达高峰。与空白对照组相比差异有显著性(P<0.05)。结论醛固酮刺激H9C2细胞后,通过与盐皮质激素受体结合后促进转化生长因子β和结缔组织生长因子的表达进而促进胶原表达。
【Objective】 To detect the effect of aldosterone on collagen expression in H9C2 cells.【Methods】 Upon the model of cultured H9C2 cells,100 nM aldosterone was used to stimulate the cells for 0,1,3,6,12,24,48 hours and detect the expression of mineral cortical receptor,TGFβ,CTGF,COLⅠand Ⅲ.【Results】 After the stimulation of aldosterone,the expression of mineral cortical receptor was increased from 3 h and reach peak at 6 h by western blot;After the stimulation of aldosterone,the expression of TGFβ and CTGF was increased from 6 h and reach peak at 12 h;the expression of COLⅠand Ⅲ was increased from 6 h and reach peak at 24 h by real-time PCR.They were significant difference compared with blank control group(P 0.05).【Conclusions】 The promoting effect of aldosterone on collagen expression in H9C2 cells was induced by the combination with mineral cortical receptor and through the increased expression of TGFβ and CTGF.
