摘要
目的通过对Aurora-B在骨肉瘤中的表达及其与远处转移关系的研究,探讨其是否参与肿瘤转移。方法收集60例经外科手术切除的骨肉瘤组织,10%甲醛固定,常规石蜡包埋。采用免疫组织化学方法检测Aurora-B在骨肉瘤组织中的表达,另取21例骨软骨瘤作为对照组。同时采用Western blot法检测Aurora-B蛋白在骨肉瘤细胞株U2-OS的表达情况。结果免疫组织化学法检测显示Aurora-B表达在细胞核。骨肉瘤组织标本中53.3%阳性表达,骨软骨瘤组织中14.3%阳性表达。骨肉瘤组织和骨软骨瘤组织中Aurora-B的表达比较差异有显著性(P<0.05),但与组织类型无相关性。在14例发生远处转移的骨肉瘤标本组织中11例Au-rora-B阳性表达;阳性表达率为78.6%明显高于未发生远处转移的标本组织中Aurora-B阳性表达率45.7%(21/46),而且有丝分裂期的骨肉瘤细胞Aurora-B染色较深。Western blot法检测同样证实Aurora-B在人骨肉瘤细胞株U2-OS中阳性表达。结论 Aurora-B极有可能参与了骨肉瘤的发生、发展和远处转移,有可能成为骨肉瘤新的诊断及分子治疗靶点。
【Objective】 To investigate the expression of Aurora-B in osteosarcoma and the relationship of the Aurora-b expression with the distant metastasis in osteosarcoma.【Methods】 60 cases of general osteosarcoma obtained at surgery were fixed in 10% formalin and routinely processed for paraffin embedding.Immunohistochemical analysis was applied to detect the expression of Aurora-B in human osteosarcoma histocyte 21 cases of osteoenchondroma samples tissue were detected for control.And Western blot was applied to detect the expression of Aurora-B in osteosarcoma cell line U2-OS.【Results】 The positive stain of Aurora-B in the nucleus.The positive expression rate of Aurora-B was 53.3% in osteosarcoma and 14.3% in osteoenchondroma respectively.Aurora-B expression was significantly higher in osteosarcoma than in osteoenchondroma(P 0.05),though it was not correlated with histological type of osteosarcoma.There were 11 cases with Aurora-B positive expression in the 14 cases with lung or oral metastasis.The positive expression rate(78.6%) was significantly higher in the cases with distant metastasis than those without metastasis(45.7%).Aurora-B expression is stronger in mitotic cells.Western blot analysis also testified Aurora-B positive expression in human osteosarcoma cell line U2-OS.【Conclusion】 Aurora-B may be involved in the development,progression and metastasis of osteosarcoma.And Aurora-B can be a new diagnostic and therapeutic target for osteosarcoma.
