消癥散对荷H_(22)移植性肝癌细胞小鼠VEGF、Cyclin-D1表达的影响 被引量:1

Effect of Xiaozheng Powder on the Expression of VEGF and Cyclin-D1 of Mice with H_(22) Transplanted Hepatocarcinoma Cells

摘要 目的观察消癥散对荷H22移植性肝癌细胞小鼠的抑瘤作用及其机制。方法选择昆明种雄性小白鼠120只,造模成功实体瘤小鼠(60只),腹水瘤小鼠(60只),空白组(10只)。随机将实体瘤小鼠和腹水瘤小鼠分为模型组,消癥散低、中、高剂量组,5-氟脲嘧啶(5-Fu)组,5-Fu+消癥散中剂量组各6组,每组10只。接种H22肝癌细胞24h后按体重给药,每日1次,连续给药10天。实体瘤组小鼠停药24h后,称重并处死动物,剥取瘤块称重,计算肿瘤生长抑制率,采用免疫组化学法检测血管内皮生长因子(VEGF)和细胞周期蛋白D1(Cyclin-D1)表达水平。记录腹水瘤组小鼠生存天数,计算生命延长率。结果消癥散对各组实体瘤小鼠有明显的抑瘤作用,与模型组比较,除消癥散低剂量组外,其余各组差异均有统计学意义(P<0.01);消癥散能够降低瘤组织中VEGF、Cyclin-D1的表达水平,与模型组相比,除消癥散低剂量组外,其余各组差异均有统计学意义(P<0.01);消癥散能够延长腹水瘤小鼠的生存期,与模型组相比,消癥散中剂量组差异有统计学意义(P<0.05),5-Fu组和5-Fu+消癥散中剂量组差异有统计学意义(P<0.01)。结论消癥散对荷H22移植性肝癌细胞小鼠有抑瘤和延长生存期的作用,抑制肿瘤生长的机理可能与下调VEGF、Cyclin-D1的表达水平有关。 Objective To observe the tumor-inhibiting effect and its mechanism of Xiaozheng Powder(Powder for dissolving masses) on mice with H22 transplanted hepatocarcinoma cells.Methods Totally 120 Kunming male mice were selected.The successfully made solid tumor mouse models were of 60 and ascites tumor mouse models were of 60,and blank group was of 10.The solid tumor mice and ascites tumor mice were randomized respectively into the model group,Xiaozheng Powder small-dosage,medium-dosage,and large-dosage groups,5-Fu group,and 5-Fu + Xiaozheng Powder medium-dosage group.Twenty-four hours after inoculation of H22 hepatocarcinoma cells,the mice were medicated according to body weight,once a day for 10 days in succession.The mice of the solid tumor group were weighted and killed 24 hours after stop of medication and their tumors were taken out and weighted for calculating the tumor growth inhibition rate,and the immunohistochemical method was applied to detect the expression of vascular endothelial growth factor(VEGF) and Cyclin-D1.The survival days of the mice of ascites tumor group were recorded to calculate their survival rate.Results Xiaozheng Powder functioned significantly to inhibit the tumors of the solid tumor mice of all groups,compared with the model group,except the Xiaozheng Powder small-dosage group,the difference of all other groups was statistically significant(P〈0.01).Xiaozheng Powder could decrease the expression of VEGF and Cyclin-D1 in the tumor,compared with the model group,except the Xiaozheng Powder small-dosage group, the difference of all other groups was statistically significant(P〈0.01). Xiaozheng Powder could prolong the survival of ascites tumor mice,compared with the model group,the difference of Xiaozheng Powder medium-dosage group was statistically significant(P〈0.05),and the difference of 5-Fu group and 5-Fu + Xiaozheng Powder medium-dosage group was statistically significant(P〈0.01).Conclusion Xiaozheng Powder has the function of inhibiting the tumor and prolonging the life of H22 transplanted hepatocarcinoma mice.The mechanism of inhibiting tumor is possibly related with its down-regulation of VEGF and Cyclin-D1 expression.
