一氧化氮对肺泡巨噬细胞致炎效应的调节作用 被引量:6

摘要 目的 探讨一氧化氮(NO)对肺泡巨噬细胞致炎效应的调节作用。方法 内毒素(LPS)刺激体外培养的小鼠肺泡巨噬细胞,观察TNFα等细胞因子释放及NO对细胞因子释放的影响。结果 LPS诱导肺泡巨噬细胞释放炎症性细胞因子的时效关系不同,肺泡巨噬细胞释放TNFα、IL-1β、IL-6的峰值时间分别为6、8、24 小时。NO合成酶抑制剂甲基异硫脲明显降低LPS诱导的NO释放,同时强烈刺激肺泡巨噬细胞释放IL-1β、IL-6,但对TNFα释放无明显影响。外源性补充NO明显抑制IL-Iβ、IL-6 释放,亦明显抑制IL-10 释放。结论 内源性及外源性NO对肺泡巨噬细胞炎症性细胞因子释放具有明显抑制作用。 Objective To observe the regulatory effects of nitric oxide (NO) on inflammatory reaction of alveolar macrophage.Methods Alveolar macrophages from mice were stimulated with LPS 10mg·L -1 and S methylisothiorea sulfate(SMT) or S nitroso N acetyl D,L pennicillamine (SNAP).Results Following LPS stimulation,TNFα,IL 1 β and IL 6 levels peaked at the 6th h,12th h and 24th h respectively in alveolar macrophages.SMT inhibited LPS induced NO release and resulted in an increase of IL 1β and IL 6 secretion in alveolar macrophages,whereas the TNFα levels remained unchanged.SNAP inhibited IL 1β and IL 6 secretion in a dose dependent manner,but exerted no effect on TNFα release.Conclusion Endogenous and exogenous NO can inhibit the LPS induced inflammatory cytokines secretion.
