摘要 目的探讨着色性干皮病互补组D(XPD)基因751位点单核苷酸多态性与吸烟因素对肺癌发生风险的影响。方法选取肺癌患者及无肺部疾患的体检人群各126例,通过问卷调查了解吸烟、性别等一般情况,聚合酶链反应-限制性片段长度多态性(PCR-RFLP)法对XPD 751Lys/Gln进行多态性分析,并比较后者与吸烟因素对肺癌发生的影响。结果 XPD 751Lys/Lys缺失的个体患肺癌的风险明显升高(调整的OR=2.83,95%CI为1.22~6.54),在表型为Lys/Gln或Gln/Gln且吸烟的个体发生肺癌的风险大大增加,校正的OR值为6.92(95%CI为1.72~20.36,P=0.003)。结论 XPD 751Lys/Lys基因的缺失与吸烟对肺癌的发生存在一定程度的交互作用。 Objective To investigate the association of xeroderma pigmentosum group D ( XPD ) single nucleotide polymorphism at 751 site and smoking on the risk of lung cancer. Methods 126 cases of lung cancer patients and another 126 cases of medical examination without lung disease were selected. General information such as smoking, sexuality and so on were found out through a questionnaire survey. XPD 751Lys/Gln polymorphism analysis was completed by polymerase chain reaction-restriction fragment length polymorphism ( PCR-RFLP ) method. The influence of the latter and smoking on occurrence of lung cancer was analyzed. Results The risk of lung cancer in individuals missing XPD 751Lys/Lys was significantly increased ( adjusted OR=2.83,95%CI was 1.22 - 6.54 ). XPD 751Lys/Gln or Gln/Gln and smoking greatly increased the risk of lung cancer (adjusted 0R=6.92,95%CI was 1.72 ~ 20.36, P=O.003 ). Conclusion There is a certain degree of interaction between XPD 751Lys/Lys gene deletion and smoking on occurrence of lung cancer.
