摘要 目的研究去甲斑蝥素对人胰腺癌CFPAC-1细胞的促凋亡活性及其与胞内[Ca2+]i浓度的关系。方法流式细胞术检测细胞凋亡;琼脂糖凝胶电泳检测断裂DNA;激光共聚焦显微镜分析细胞内游离Ca2+浓度变化。结果 CFPAC-1细胞经去甲斑蝥素作用24h后,细胞凋亡率明显上升,且呈剂量依赖性,其基因组DNA在琼脂糖凝胶电泳中呈典型的梯状带型;CFPAC-1细胞内游离Ca2+浓度进行性升高。结论去甲斑蝥素触发的CFPAC-1细胞凋亡与胞内游离Ca2+浓度升高密切相关。 Objective To study the proapoptotic effect of Norcantharidin ( NCTD ) on the CFPAC-1 Cells and its relationship to the changes of intracellular [Ca^2+]i. Methods Flow cytometry were used to evaluate HepG2 cells apoptosis, agarose gel electrophoresis was used to detected DNA fragmentation; and the changes of the concentration of intracellular [Ca^2+]i analyzed by confocal laser scanning microscope ( CLSM ). Results After treated with NCTD, a dose-dependent apoptosis was also observed by ow cytometery analysis, a characteristic DNA "ladder" pattern was found in agarose gel electrophoresis, and the concentration of intracellular Ca^2+ in target cells was progressively elevated. Conclusion NCTD induced apoptosis in CFPAC-1 cells, appears to be closely associated with the increase in the concentration of intracellular [Ca^2+]i.
