摘要
目的探讨RASSF1A基因对人胃癌SGC7901细胞NF-κB活性的影响。方法电泳迁移实验分析RASSF1A基因转染前后SGC7901细胞NF-κB活性的变化。蛋白印迹和免疫细胞化学检测RASSF1A基因转染前后SGC7901细胞P65蛋白的表达。结果与未转染组和转染空白载体组比较,转染RASSF1A基因组SGC7901细胞的NF-κB活性明显降低,P65蛋白的表达明显减少。结论 RASSF1A基因可能通过抑制P65的表达、降低NF-κB活性抑制从而抑制胃癌SGC7901细胞生长。
Objective To study the effects of RASSF1A gene on activity of NF-κB in human gastric carcinoma cell line SGC7901.Methods NF-κB activity was measured by EMSA(electrophoretic mobility shift assay).Expression of P65 protein was detected by Western blotting and immunocytochemistry.Results Compared with the non-transfected and vector transfected cells,NF-κB activity of RASSF1A-SGC7901 cells was lower.The expression of P65 protein in RASSF1A-SGC7901 cells was lower than that of non-transfected and vector transfected cells.Conclusions RASSF1A gene could suppress the activity of NF-κB in SGC7901 cells.
