非小细胞肺癌中DR5和DcR1的表达与临床病理特征及预后的关系 被引量:1

Expression of DR5 and DcR1 in human non-small cell lung cancer and its relationship with clinical significance and prognosis

摘要 目的探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)死亡受体5(DR5)、诱捕受体1(DcR1)在非小细胞肺癌(NSCLC)组织中的表达及其与临床病理特征、预后的关系。方法采用免疫组织化学法检测45例NSCLC组织及其相应的42例癌旁组织中DR5、DcR1蛋白的表达。结果 DR5在45例NSCLC组织中表达率为86.7%(39/45),其中高表达率为53.3%(24/45);在42例癌旁组织中为76.2%(32/42),高表达率为11.9%(5/42),两组比较差异有统计学意义(P〈0.01);DcR1在NSCLC组织中表达率为28.9%(13/45),而在对应癌旁组织为71.4%(30/42),差异有统计学意义(P〈0.01)。DR5的表达水平与临床分期有关,Ⅰ~Ⅱ期的表达量显著高于Ⅲ期(P〈0.05)。DR5、DcR1在NSCLC组织中的表达与性别、年龄、肿瘤大小及预后均无关。结论 DR5、DcR1在NSCLC组织及癌旁组织中均有表达,但存在表达量的差异。DR5的高表达可能在调控NSCLC的凋亡中发挥重要作用。 Objective To investigate the expression of tumor necrosis factor-related poptosis-inducing ligand(TRAIL) death receptor 5(DR5) and decoy receptor 1(DcR1) in human non-small cell lung cancer(NSCLC) and the relationship with clinical significance and prognosis.Methods Immunohistochemistry was used to detect the expression of DR5 and DcR1 in 45 cases of NSCLC and 42 cases of non-cancerous adjacent lung tissues and their clinical pathological parameters was analyzed.Results The expression rate of DR5 in NSCLC was 86.7%,and the high expression rate was 53.3%,while that in non-cancerous adjacent tissues was 76.2% and 11.9% respectively,showing statistic significance(P0.01).The expression rate of DcR1 in NSCLC was 28.9%,which was significantly lower in non-cancerous adjacent tissues(76.2%).The expression level of DR5 in NSCLC tissue was correlated with clinical stage,and the expression level of stage Ⅲ was significantly lower than that of stage Ⅰ and Ⅱ(P0.05),while the expression of DR5 and DcR1 in NSCLC tissue didn't relate to gender,age,the size of tumor and prognosis.Conclusion TRAIL receptors express in both NSCLC and non-cancerous adjacent lung tissues,but the difference of expression levels exists.The high expression level of DR5 might play an important role in TRAIL-induced apoptosis of NSCLC.
