摘要
目的:本实验对肉桂酸锗的毒副作用进行了初步探讨。方法:采用急性毒性试验计算出药物对昆明小鼠的半数致死剂量(Median lethal dose,LD50)和最大耐受量(Maximum tolerance dose,MTD);通过药理试验连续观察小鼠肢体活动和行为改变等指标,检测血液生化指标和血常规,给药7 d后处死小鼠称体重,尸解并取出各脏器,计算脾重指数(Spleen index,SI)和胸腺指数(Thymus index,TI);经HE染色后光镜下观察各个脏器病理形态变化。结果:无法计算出LD50,说明药物毒性甚小,动物MTD可达到1.2 g/kg体重;各组血液生化指标和血液常规差异无统计学意义,说明药物无明显肝肾毒性,对血液常规亦无明显影响;各组小鼠体重、SI和TI差异无统计学意义,说明药物对小鼠免疫功能短期内没有明显影响;给药组与对照组比较,各脏器镜下未见明显药物损伤的病理形态变化。结论:肉桂酸锗治疗剂量在短期内无明显毒副作用,这为进一步的抗肿瘤活性研究提供了实验依据。
Objective:To study the toxic effects of cinnamon acid germanium in the experiment. Methods: Acute toxicity tests were adopted to calculate the median lethal dose and the maximum tolerated dose of the drugs; and to check mice' s biochemical indicators. The various organs were drawn out after mice were killed by pharmacological test, and the spleen and thymus weight indexes were calculated; The organs' pathological changes were observed under ordinary microscope after HE staining. Re,s'~ts:LDso could not be calculated, which showed cinnamon acid germanium had little toxicity. The maximum tolerated dose of mice could reached 1.2 g/kg weight. Mice' s biochemical indicators had no statistical significance. The spleen and thymus weight indexes in treated group have no statistical significance,This indicated the drags' immune influence in mice had no significant effect in the short term.Compared with normal saline group and cyclopho-sphamide group,the organs in treated group showed no obvious pathological changes under ordinary microscope. Conclusion:The experiment shows that cinnamon acid germanium compounds have no apparent toxic effects. This can provide some experimental evidence for further anti-tumor activity researches and applications.
