β-防御素-2、IFN-γ和NF-κB p65在寻常型银屑病皮损中表达的研究 被引量:7

摘要 目的研究β-防御素-2(HBD-2)、IFN-γ、NF-κB p65在寻常型银屑病患者皮损中的表达及其与银屑病严重程度的关系。方法采用SP免疫组化法检测45例银屑病患者皮损组织、45例银屑病患者非皮损组织及15例正常皮肤组织石蜡标本中HBD-2、IFN-γ、NF-κB p65的表达,对其在皮损中的表达进行相关性分析,并将结果与PASI评分进行相关性分析。结果与正常人皮肤组织和银屑病患者非皮损区相比,银屑病患者皮损区中HBD-2、IFN-γ、NF-κB p65的表达明显上调,而非皮损区HBD-2、IFN-γ的表达也高于正常人皮肤组织,差异均有统计学意义(P<0.05)。银屑病患者皮损区HBD-2、IFN-γ、NF-κB p65的表达水平与PASI评分之间均存在正相关(P<0.05)。银屑病患者皮损区HBD-2与IFN-γ,IFN-γ与NF-κB p65,HBD-2与NF-κB p65表达水平之间均存在正相关(P<0.05)。结论 HBD-2,IFN-γ,NF-κB p65在寻常型银屑病病中的表达水平均升高,可能共同参与了银屑病的发病过程。 To investigate the expressions of β - defensin - 2 ( HBD - 2 ) , interferon - gamma ( IFN - γ) , and nuclear - factor - κB p65 (NF - κBp65) in the epidemis of psoriasis vulgaris and study their correlation with disease severity in psoriatic pa- tients. Methods Forty - five samples of lesional psoriatic skin,forty - five samples of nonlesional psoriatic skin and fifteen samples of normal skin were analyzed by Elivision immunohistochemical technique for the expressions of HBD - 2 ,IFN - γand NF - κB p65. Correla- tion analysis was performed among the expressions of the three factors as well as between the expressions and disease severity. Results Compared with those of normal epidermis and nonlesional psoriatic epidermis, HBD-2,IFN-γ,and NF-κB p65 were more highly expressed in lesional epidermis from patients with psoriasis (all P 〈 O.05 ). In nonlesional psoriatic epidermis, HBD-2, IFN-γ expression was also upregulated compared with that of normal epidermis ( P 〈 0.05 ). The expressions of HBD-2,IFN-γand NF - κBp65 in lesional epidermis positively correlated individually with the psoriasis area and severity index(PASI) scores in patients (all P 〈 0.05 ). Positive correlation was also found between the expressions of any two factors( HBD -2, IFN-γor NF - KB p65 ) in lesional epidermis( all P 〈 0. 05). Conclusion The upregulation of HBD - 2,IFN - γ and NF - κB p65 expressions in psoriatic lesions may be involved in the pathogenesis of psoriasis.
