摘要
提取介质中加入25% β巯基乙醇, 能有效去除次生物质对木麻黄基因组DNA 提取的影响. 用新建方法所提DNA 产率比用前人方法提的高出08 倍左右. 鉴定结果表明: 鲜小枝、干小枝的DNA 样品皆为48kb 左右, 适于限制性酶切和RAPD 反应; 同时发现同一株树的鲜小枝、干小枝,基因组DNA
The extraction medium,which mixed with 2.5%β mercaptoethanol,can eliminate the secondary materials' effections to extracting gemone DNA from Casuarina equisetifolia effectively.The yields of genome DNA extract with the new method are 1 8 times as many as those of the DNA extracted with the former method.The result of characterization shows that the molecular weights of genome DNA from fresh twigs and dry twigs are 48kb,which are all suitable for digesting with restrictive enzyme and RAPD analysis.In addition, DNA templates extracted from fresh twigs and dry twigs produce the same RAPD result.
