摘要
以欧洲大黄种子为材料,进行了组织培养和快速繁殖技术的研究。试验结果表明:欧洲大黄种子获取无菌实生苗的较好处理方式是将种子剥壳,胚根垂直向下;种子进行无菌萌发用0.1%的氯化汞溶液消毒时,消毒时间以8分钟为宜;子叶是对愈伤诱导的最佳外植体;茎芽诱导的最佳培养基是MS+BA1.0mg/l+NAA1.0mg/l;1/2MS是适宜的生根培养基。
With the material of Rheum rhap onticurn L.,tissue culture and rapid reproduction technology are used in this research.The results of this research show that a better way to get a sterile seedling of Rheum rhap onticurn L.is to husk the seed and let the radicle vertically downward;in sterile germinating the seed,it should be disinfected with 0.1% mercuric chloride solution for 8 minutes.Cotyledon is the optimal explant for callus inductions;the optimal culture medium for stem eye induction is MS＋BA1.0mg/l＋NAA1.0mg/l;1/2MS is the proper culture medium of taking root.
