家兔克隆骨髓基质细胞的体外培养

摘要 目的探讨家兔骨髓基质细胞在体外传代的培养。方法取家兔股骨骨髓,分离骨髓基质细胞,于1640培养体系中培养,之后固定计数。结果在2~6×106个/cm2范围内,其接种细胞数与所形成的克隆集落(CFU-F)数呈良好线性关系。随着培养时间的延长,克隆集落也增多,变大。3周龄家兔的造血基质祖细胞较6周龄者为多。结论基质祖细胞CFU-F呈非均质性的,可能存在不同亚群。 Objective To investigate the growing variation of the culture of colonies of rabbit bone marrow stomal cell in vitro Methods Extracted rabbit bone marrow stomal cell,then cutlured these cell isolated in 1640 cultre system of CO 2 incubator Count the colony after fixed Results There was a clearly linear relationship between the number of implanted cells and the formed colonies in a some concentration(2~6×10 6 /cm 2),and the colonies of bone marrow stromal cell were increasing with the time extending.The CFU F of the three weeks rabbit were more than that of the six weeks rabbit Conclusion The CFU F are nonhomogenous,and might be heterogeneous subgroups.
