肿瘤转移相关因子RhoGDI2蛋白在肺癌95D细胞侵袭转移中的作用

Role of RhoGDI2 protein on invasion and metastasis of human lung cancer 95D cell line

目的研究肿瘤转移相关因子RhoGDI2蛋白在肺癌95D细胞侵袭转移中的作用。方法应用细胞培养技术培养95D细胞,分别10,30μg/L不同浓度rhHGF作用95D细胞48 h后收集细胞沉淀,利用蛋白印迹方法检测RhoGDI2蛋白表达情况。结果无血清培养的95D细胞中RhoGDI2蛋白的表达,高于正常10%胎牛血清培养的细胞组,而10%胎牛血清培养的细胞组与10ng/mL rhHGF组RhoGDI2蛋白的表达水平相当,但30ng/mL rhHGF组RhoGDI2蛋白表达明显降低。结论促进肺癌95D细胞生长、侵袭转移的因素与RhoGDI2的表达成负相关。

【Objective】 To study the role of RhoGDI2 on invasion and metastasis of 95D human lung cancer cells.【Methods】 95D cells of human lung cancer were cultured by cell culture technique.95D cells were incubated with 10,30 μg/mL rhHGF for 48 h.Western Bolt assay was used to evaluate the expressions of RhoGDI2 protein.【Results】 In serum-free culture of 95D cells,the expression of RhoGDI2 is higher than that in 95D cells cultured in the presence of 10% fetal bovine serum.10ng/mL rhHGF treatment did not affect the expression of RhoGDI2 compared to that in 95D cell culture in 10% FBS,whereas 30ng/mL rhHGF significantly reduced it.【Conclusion】 The expression of RhoGDI2 is negatively related with the invasion and metastasis of lung cancer 95D cell line.
