丹参酮ⅡA磺酸钠对TGF-β1诱导肾小管上皮细胞PAI-1表达的影响 被引量:5

摘要 目的探讨丹参酮ⅡA磺酸钠对TGF-β1诱导的人肾小管上皮细胞纤溶酶原激活物抑制因子1(PAI-1)表达的影响,为防治肾间质纤维化提供有效的途径。方法人近曲小管上皮细胞株(HKC-8),予TGF-β1(2ug/mL)刺激48h,同时给予丹参酮ⅡA磺酸钠(终浓度分别为0.5、1.0、2.0、4.0mg/mL)进行干预,采用RT-PCR、western blot等方法检测PAI-1的表达。结果与对照组比较,TGF-β1明显上调PAI-1mRNA及蛋白的表达(P<0.01),丹参酮ⅡA磺酸钠以浓度依赖方式抑制TGF-β1介导的PAI-1上调(P<0.05)。结论丹参酮ⅡA磺酸钠能逆转TGF-β1诱导的人肾小管上皮PAI-1的表达,因而具有防治肾间质纤维化的作用。 【Objective】 To investigate the protective effects of Sulfontanshinone ⅡA Sodium(STS)on the expression of plasminogen activator Inhibitor-1(PAI-1) induced by transforming growth factor-β1(TGF-β1)in human renal tubular epithelial cells.【Methods】 Human kidney proximal tubular cell line(HKC-8)was used as the proximal tubular cell model.Cells were treated with TGF-β1(2ng/mL)plus STS(0.5、1.0、2.0、4.0mg/mL) for 48 hours,the expressions of PAI-1mRNA and protein were measured by RT-PCR、western blot.【Results】 Our results revealed that TGF-β1can up-regulate the expression of PAI-1 mRNA and protein(P0.01),STS can inhibit the expression of PAI-1 mRNA and protein which induced by TGF-β1 in HKC-8 cells in a dose-dependent manner(P0.05).【Conclusion】 These observations strongly suggest that STS is a potent inhibitor of TGF-β1-induced PAI-1 and may be a promising agent for treating renal interstitial fibrosis.
