转化生长因子-β对骺软骨细胞增殖周期的作用 被引量:4

摘要 采用鸡胚股骨无血清培养和流式细胞仪,对经转化生长因子β( T G Fβ)作用后的骺软骨细胞进行检测。结果显示:24 小时时,0.11、0.33 和1.00ng 实验组的细胞增殖指数( P I)均低于对照组,而3.00ng 组则高于对照组( P< 0.001);48 小时时,除0.11ng 组外,其余各实验组 P I均低于对照组( P< 0.001);96 小时时,各实验组 P I均大于对照组,其中0.33ng 组 P I最大( P< 0.001)。上述结果表明,不同剂量和不同作用时间的 T G Fβ对骺软骨细胞增殖周期均有较大的影响,其总的趋势是, T G Fβ抑制细胞的增殖,然后随着作用时间的延长,抑制作用转变为促进细胞增殖作用。 In order to investigate the effect of transforming growth factor β(TGF β) on chondrocyte proliferation cycle, we used flow cytometry to assess the epiphyseal chondrocyte of embryonic chick femora which was cultured in free serum media and treated with TGF β.The results showed that after treated with TGF β for 24h, the proliferation indexes(PIs) of experimental groups(0.11ng, 0.33ng, 1.00ng/ml) were lower than the PIs of the control groups, but the PI of 3.00ng/ml group was higher than that of the control group ( P<0.001) . After treated for 48h, except the 0.11ng/ml group, the PIs of experimental groups were lower than those of the control ( P<0.001) . And after treated for 96h, the PIs of all groups were higher than those of the control; the PI of the 0.33ng/ml group was the highest ( P<0.001) . These suggest that after treated in varied time and dosage, the effect of TGF β on epiphyseal chondrocyte is obvious, and TGF β on epiphyseal chondrocyte is obvious, and TGF β can inhibit chondrocyte proliferation first, and then its effect on chondrocyte can transform from inhibition into proliferation.
