摘要
胆盐水解酶主要是微生物在生长和繁殖过程中所产生的代谢产物;具有降低胆固醇的功能。克隆植物乳杆菌中的胆盐水解酶基因并对其序列进行分析。从植物乳杆菌中提取基因组DNA作为模板,根据Genbank上胆盐水解酶全长基因序列(EU477374)设计一对特异性引物,采用PCR扩增技术得到植物乳杆菌中胆盐水解酶基因并对其进行测序。结果表明:克隆的胆盐水解酶结构基因全长为995 bp,将其核苷酸序与Genbank(EU477374)上已报道的胆盐水解酶结构基因序列进行比对,同源性高达99.3%。结论:成功克隆胆盐水解酶结构基因,这为胆盐水解酶的进一步表达奠定了基础。
The BSH gene of Lactobacillus plantarum was cloned and sequence analyzed.Extract the genome DNA of Lactobacillus plantarum.According to the complete gene sequence of BSH in Genbank(EU477374),a pair of specific primer was designed,BSH gene of Lactobacillus plantrum were obtained by PCR amplify and predicted the sequence.Result indicate: The total length of cloned BSH gene was 995 bp;The homology between the predicted amino acid sequence and the EU477374 was 99.3%.Conclusion: BSH gene was successfully cloned which may lay the foundation for expression of BSH.
