摘要
利用亨盖特厌氧操作技术,从石家庄污水处理厂活性污泥中分离出一株产甲烷古菌QDW。该菌株为不规则球形,直径1~2μm,不产芽孢、革兰氏阴性、不运动、极端严格厌氧。最适生长温度为37℃,最适pH为7.0~7.5,最适盐浓度〈0.1 mol/L,以甲酸钠、乙酸钠、乙二醇、H2/CO2为底物产甲烷,不能利用甲醇、丙二醇和甲基胺。对青霉素、链霉素、罗红霉素、利福平有抗性,对氯霉素、四环素、新霉素敏感。经生理、生化及16S rDNA分析,确定该菌株为甲烷袋状菌属(Methanoculleus sp).。
An improved Hungate roll-tube technique was used to isolate a strain of methanogen from a sewage treatment plant of Shijiazhuang,Hebei Province.The strain QDW is obligate anaerobes,with the cells of organism as irregular non-motile cocci,1~2 μm in diameter,non-producing spores and Gram-negative.The optimum growth temperature was 37 ℃,the fastest growth occurred within a pH range of 7.0~7.5 and optimum NaCl concentration for growth less than 0.1 mol/L.H2/CO2,Na-formate,Na-acetate,glycol were used as substrate for methane production.Neither growth nor methane production were observed with monomethylamine,dimethylamine methanol,methanol and trimethylene glycol as substrates,which were resistant to Penicillin,Streptomycin,Roxithromycin and Rifampicin,while sensitive to tetracycline,chloramphenicol and neomycin.The physiological biochemical characteristics and 16S rDNA analysis of the strain indicated that the strain is a member of genus Methanoculleus sp.
