摘要
目的:探讨EPHB6对非小细胞癌(NSCLC)转移的抑制作用。方法:采用体外的黏附和迁移实验以及小鼠体内转移模型来评价EPHB6在NSCLC转移中的作用。过表达和RNAi干涉技术用于对EPHB6生物学功能的分析。结果:恢复EPHB6在肺腺癌细胞中的表达可增强细胞的黏附能力、降低细胞迁移能力。在患肺癌的非肥胖糖尿病/严重联合免疫缺陷(NOD/SCID)小鼠中,EPHB6的重新表达对肿瘤转移有明显抑制作用。结论:EPHB6在非小细胞肺癌中是一种肿瘤转移抑制基因。
Objective:To explore EPHB6 inhibiting the metastasis of non-small cell lung cancer(NSCLC).Methods:Metastatic potential of EPHB6 was assessed by both in vitro experiments of adhesion and migration and in vivo metastasis mouse model.Over expression and RNAi amethods were used to analyse the biological functions of EPHB6.Results:Restoration of EPHB6 expression in lung adenocarcinoma cells increased adhesion and decreased migration.Reexpression of EPHB6 in lung cancer cells almost entirely abolished metastasis formation in NOD/SCID mice.Conclusion:Taken together,these analyses demonstrate that EPHB6 is a metastasis inhibitory gene in non-small cell lung cancer.
