摘要
目的观察三七有效组分Rx对兔动脉粥样硬化碱性细胞生长因子(bFGF)表达的影响,以探讨其防治动脉粥样硬化(AS)的作用机制。方法通过喂高脂饲料建立兔动脉粥样硬化模型。48只新西兰兔随机分正常组、模型组、三七总皂苷(PNS)组、三七有效组分Rx高剂量组、三七有效组分Rx低剂量组、卡托普利组,并进行相应干预12周。观察各组主动脉形态学变化;检测各组血浆一氧化氮(NO)、脂质过氧化物(LPO)、超氧化物歧化酶(SOD)和bFGF表达。结果与模型组比较,各药物组主动脉粥样硬化程度均有减轻;血浆NO水平和SOD水平升高(P<0.01),血浆LPO和bFGF表达降低(P<0.01);Rx高剂量组与PNS组、Rx低剂量组比较有显著性差异(P<0.05)。结论三七有效组分Rx通过升高NO、提高SOD活性,增强抗脂质过氧化和抑制bFGF表达,起到防治动脉粥样硬化作用。
Objective To observe the effects of the effective fractions(Rx) of Panax Notoginseng on expression of basic fibroblast growth factor(bFGF) in rabbit atherosclerosis(AS),in an attempt to study its mechanisms.Methods AS model was established by feeding high-lipid fodder.Forty-eight rabbits were divided into six groups: normal group,model group,PNS group(by total Saponins of Panax Notoginseng),high-dose Rx group,low-dose Rx group,and Captopril group,with a course of 12 weeks.Some segments of aorto were taken for pathologic morphology.The levels of nitrogen oxide(NO),lipid peroxide(LPO),and speroxide dismutase(SOD) were measured;and the expression of bFGF was detected by immunochemistry.Results Compared with the model group,the atherosclerosis lesions were relieved obviously in all intervention groups;the levels of plasma NO and SOD were increased(P〈0.01),the level of LPO and bFGF expression were decreased(P〈0.01);there were significant differences in all indicators between high-dose Rx group and PNS and low-dose Rx groups(P〈0.05).ConclusionThe effective fractions of Panax Notoginseng can fight atherosclerosis by increasing the levels of NO and SOD,reducing LPO and bFGF expression.
