摘要
采用冬绿金丝桃带腋芽的茎段作为外植体进行离体培养试验,结果表明:在1/2MS培养基上,当NAA浓度为0.1 mg/L,BA浓度取0.4 mg/L时,冬绿金丝桃的芽诱导率高达3.43,去除顶端是促进芽发生的有效途径;在1/2MS+IBA 0.1 mg/L培养基上生根效果明显,生根率达到95%,移栽成活率在80%以上。建立了高效的冬绿金丝桃离体培养快繁技术体系并进行了工厂化生产,成功地生产出株性一致的冬绿金丝桃组培苗30万株,为推广冬绿金丝桃良种奠定了基础。
The study on tissue culture and in vitro rapid propagation of Hypericum calycinum was developed using nodal segments as explants.The results showed that the efficient medium for the first and multiplated culture was 1/2 MS,when the concentration of NAA and BA was 0.1 mg/L and 0.4 mg/L,in which the average multiplication rate of shoots was reached to 3.43.And the tip cutting was a good method for shoots regeneration.In the medium of 1/2 MS+IBA 0.1 mg/L,the rooting rate was about 95% after one month and the transplant survival rate was 80%.The technological processes of H.calycinum for the commercialized production was established and 300 000 plantlets were produced.
