知母中具抑瘤活性的芒果苷的微波辅助提取工艺 被引量:7

摘要 目的优化知母中微波辅助提取芒果苷的工艺条件,研究其对人乳腺癌细胞株MDA-MB-231的增殖抑制作用。方法采用单因素试验,考察乙醇浓度、提取时间及固液比对提取得率的影响。以四甲基偶氮唑盐微量酶反应比色法(MTT法)观察不同浓度芒果苷对MDA-MB-231细胞生长的影响。结果微波辅助提取芒果苷的最佳工艺条件为乙醇浓度30%,提取时间60 s,固液比1∶40(g/mL),在此条件下,芒果苷的得率为1.82%。MTT法显示,不同浓度的芒果苷对MDA-MB-231细胞均有抑制作用,其中1、5、10μmol/L浓度芒果苷对MDA-MB-231细胞抑制作用最为显著,其24h抑制率分别为20.55%、22.26%和22.94%。结论微波辅助提取省时,操作简单,收率高;芒果苷能明显抑制人乳腺癌细胞株MDA-MB-231的增殖。 OBJECTIVE To investigate the best extraction condition of microwave extraction of mangiferin in Anemarrhena asphodeloides Bge,study the effects of proliferation inhibition of mangiferin on MDA-MB-231 cells of breast cancer.METHODS Monofactorial experiment was carried out to investigate three influential factors,including alcohol concentration,extracting time and solid-liquid ratio,and their effect on extraction rate.The effect of different concentrations of mangiferin on the proliferation of MDA-MB-231 cells was examined by MTT assay.RESULTS It was showed that the best extraction condition was to have alcohol concentration at 30%,extraction time for 6 min and the solid-liquid ratio at 1∶40(g/mL),in which the yield of manhiferin extracts in Anemarrhena asphodeloidge Bge was 1.82%.MTT assays showed that different concentrations of mangiferin can inhibit the MDA-MB-231 cells proliferation,the inhibition of MDA-MB-231 cells was 20.55%,22.26% and 22.94% respectively with(1μmol/L,5μmol/L,10μmol/L) mangiferin treatment for 24 hours.CONCLUSION Microwave extraction is time-saving,easy to operate and highly efficient.The mangiferin can significantly inhibit the proliferation of MDA-MB-231 cells.
