摘要
目的研究苯[α]芘对人B淋巴细胞株2个短串联重复序列位点的遗传效应,探讨人细胞体外系统的遗传稳定性和敏感性。方法采用聚合酶链反应(PCR)分析人B淋巴细胞株2个短串联重复序列位点FGA和D18S379的扩增产物。结果经500、50、5和0.5μmol/L苯[α]芘分别处理细胞株,未发现FGA和D18S379位点发生杂合性丢失,未发现这2个位点出现等位获得,也没有发现重复单位增加。结论在2个目标位点FGA和D18S379没有发生遗传多态性变化。
[Objective]To study the genetic effect of benzo[α]pyrene on two short tandem repeat loci in human B lymphoblast cell line,explore the genetic stability and sensitivity of human cell culture system. [Methods]The amplification products of two short tandem repeat loci FGA and D18S379 in human B lymphoblast cell line were analyzed by PCR. [Results]The cell lines were respectively treated with 500,50,5 and 0.5 μmol/L benzo[α]pyrene,loss of heterozygosity,allele gain and repeat unite amplification were not shown in FGA and D18S379. [Conclusion]The results suggest an absence of changes in genetic polymorphism of two target loci FGA and D18S379.
