碱性成纤维细胞生长因子和胰岛素样生长因子-I及其协同作用对牛晶体上皮细胞增殖的影响 被引量:7

Effect of bFGF, IGF Ⅰ and their synergy on the mitogenesis of bovine lens epithelial cells

摘要 目的 研究碱性成纤维细胞生长因子(basicfibroblastgrowthfactor,bFGF)、胰岛素样生长因子I(insulinlike growth factorI,IGFⅠ) 及二者的共同作用对体外牛晶体上皮细胞(bovine lensepithelialcell,BLEC)增殖的影响。方法 BLEC原代培养,取第4 代细胞种入24 孔板,加入不同浓度的bFGF、IGFⅠ、bFGF+ 20 μg/LIGFⅠ,20 h 后加入3HTDR,16 h 后作液闪计数。结果 一定浓度的bFGF(1~100 μg/L) 、IGFⅠ(20~100 μg/L) 在体外有促进BLEC 增殖的作用( P< 0-01 ) ,20 μg/L的IGFⅠ可增加bFGF的促BLEC增殖作用。结论 生长因子及它们之间的协同作用在促进晶体上皮细胞异常增殖的过程中起重要作用。 Objective To assess the effect of basic fibroblast growth factor (bFGF), insulin like growth factor Ⅰ(IGF Ⅰ) and their synergy on the mitosis of bovine lens epithelial cells (BLECs) Methods BLECs were cultured in vitro The fourth passage cells were inoculated into 24 well tissue culture plates, treated with different concentrations of bFGF, IGF Ⅰ, and bFGF+20 μg/L IGF Ⅰ for 20 hours, 3H thymidine was added for 16 hours, then the radioactivity was determined by liquid scintillation counting Results Either bFGF (1~100 μg/L) or IGF Ⅰ (20~100 μg/L) increases mitosis of BLEC significantly, and 20 μg/L IGF Ⅰ may increase bFGF induced mitogenesis Conclusion Growth factor and their synergic effect may play an important role in lens epithelial cell proliferation
