Effects of ISP on the expression of rat endothelin gene

目的 探讨异丙肾上腺素（ＩＳＰ）对内皮素基因表达的影响。方法 通过ＩＳＰ心肌损伤模型，用放免及分子生物学的方法测定血浆内皮素－１（ＥＴ－１）放免活性及心肌和主动脉 ＥＴ－１ｍ ＲＮＡ 的表达。结果 ＩＳＰ致血浆中ＥＴ－１ 放免活性明显升高，使心肌及主动脉ＥＴ－１ ｍ ＲＮＡ 高表达。结论 表明ＩＳＰ使血管内皮细胞ＥＴ－１ ｍ ＲＮＡ 高表达，内皮素分泌增多可能是其致心肌损伤的一个重要原因。

Objective The relationship between isoproterenol and the rat endothelin gene expression had been studied in this article.Methods In ISP myocardial damage model,the plasma radio immuno activity of ET 1 and ET 1 mRNA expression in myocardium and aorta were determined by means of radio immuno logy and molecular biology techniques.Results ISP can increase the radio immuno activity in rat plasma and induce high expression of ET 1 mRNA in myocardium and aorta.Conclusions The results revealled that ISP can induce ET 1 mRNA high expression in endothelial cells.The increased ET 1 secretion may be an important mechanism in ISP induced myocardial damage.
