血管内皮生长因子表达和肝癌细胞周期启动的关系

THE RELATIONSHIP BETWEEN THE EXPRESSION OF VASCULAR ENDOTHELIAL GROWTH FACTOR AND INITIATION OF CELL CYCLE IN HEPATOMA CELL LINE HEP G_2

为了探讨血管内皮生长因子（ ＶＥＧＦ） 表达与肝癌细胞增殖的联系。采用同步化环境Ｇ０ 期的Ｈｅｐ Ｇ２ 细胞用ＴＰＡ 诱导启动细胞周期并用ｃｊｕｎ 反义寡核苷酸阻断细胞周期，观察ＶＥＧＦ 表达变化。结果显示： Ｇ０ 期Ｈｅｐ Ｇ２ 不表达ＶＥＧＦ，伴随细胞周期启动ＶＥＧＦ 表达，ｃｊｕｎ 反义寡核苷酸在使Ｈｅｐ Ｇ２ 细胞周期停滞于Ｇ０／Ｇ１ 期的同时使ＶＥＧＦ 表达下调。由此表明： 在Ｈｅｐ Ｇ２ 细胞株ＶＥＧＦ 表达和细胞周期启动联动，提示二者受同一信号通路调节，ＶＥＧＦ 表达有可能成为肝癌细胞增殖的标志物。

For research the relationship between the expression of vascular endothelial growth factor (VEGF) and the cell proliferation. The expression of VEGF was evaluated while the cell cycle of hepatoma cell line Hep G 2, which was synchronized at G 0 phase with serum deprivation, and reinitiated with TPA and blocked with antisense oligonucleotides of c jun. Results: Hep G 2 cell did not express VEGF at G 0 phase. TPA could induce the expression of VEGF as well as initiation of cell cycle. The antisense oligonucleotides of c jun could prohibit the expression of VEGF and arrest the cell cycle at G 0 phase. Conclusion: The fact that the expression of VEGF accompanies the initiation of cell cycle suggests that they be regulated by the same singnal pathway, the expression of VEGF may be a marker indicating the proliferation of hepatoma cells.
