摘要
目的:探讨江苏汉族人群脂联素通路基因多态性和环境的联合作用与2型糖尿病的关系。方法:采用病例对照研究,收集新诊断T2DM病例(1105例)和健康对照(1107例)的临床和流行病学相关资料。利用Real-time PCR联合Taqman-MGB探针方法对19个多态性位点进行基因分型。结果:显性遗传模型的单因素Logistic回归分析发现ADIPOQ基因rs7649121、PPARγrs3856806基因型频率在病例与对照组的分布差异有统计学意义(P<0.05)。多因素Logistic回归分析显示糖尿病家族史、高血压史、腰围、血清脂联素浓度、吸烟、饮酒、ADIPOQ rs7649121、rs12495941,PPARγrs3856806等因素与T2DM的发病风险存在统计学关联(P<0.05)。结论:脂联素通路多态性与环境联合作用对于T2DM遗传易感性可能具有一定的影响。
Objective:To explore the relationship between the genetic variants in the adiponectin pathway jointed with the environment and type 2 diabetes risk in a Jiangsu Han population.Methods: Epidemiology questionnaires and physical examinations were performed to gain the healthy status and biochemistry markers from first-diagnosed type 2 diabetes patients(n=1 105) and controls(n=1 107).The genotypes were tested with Real-time PCR and Taqman-MGB probe assays.Results: The polymorphisms of ADIPOQrs7649121 and PPARγrs3856806 showed significant differences between type 2 diabetes cases and controls in the dominant genetic model by univariate logistic regression analysis(P0.05).With multiple logistic regression analysis,diabetes mellitus family history,hypertension,waist,serum adiponectin level,smoking,do no drink alcohol,rs7649121,rs3856806 and rs12495941 were associated with T2DM(all P0.05).Conclusion: The effect of the genetic variants in the adiponectin pathway jointed with the environment may play a role in the susceptibility of type 2 diabetes.
