摘要
以MS为基本培养基,通过与植物生长调节剂结合形成不同的组合培养基,采用野生的魔芋(Amorphophalms konjac)块茎为外植体,经过严格的消毒处理后用于试验。结果显示,在MS+1.0mg/L 6-BA+0.5mg/L NAA的培养基上容易诱导愈伤组织且效果好,MS+1.0mg/L 6-BA+1.0mg/LNAA适合做愈伤组织的增殖培养基,在MS+2.0mg/L 6-BA+0.5mg/L NAA的培养基上容易诱导出不定芽,在1/2MS+1.0mg/L 6-BA+1.0mg/L NAA的培养基上容易诱导出不定根;在愈伤组织的褐化上,采用降低无机盐和激素浓度、合理使用植物生长调节剂以及控制光照,可取得良好效果。
Different combinations of culture medium were made through adding different plant growth regulator with different concentration into MS which is a basic culture medium.Through strict disinfection,explants that come from the tubers of wild konjak were used to experiment.The results showed that on the culture medium composed of MS,1.0mg/L 6-BA,and 0.5mg/L NAA callus is prone to inducement and may get a good effect;the culture medium consisting of MS,1.0mg/L 6-BA and 1.0mg/L NAA,is suitable for callus proliferation;the adventitious buds and roots are liable to abduction respectively at the culture medium composed of MS,2.0mg/L 6-BA and 0.5mg/L NAA,and at the culture medium consisting of 1/2 MS,1.0mg/L 6-BA and 1.0mg/L NAA;adopting appropriate measures such as inorganic salt and hormone with a low concentration,reasonable use of plant growth regulator,as well as illumination control,may get a good effect in browning improvement of konjak callus.
