摘要
为提高羊软骨硫酸软骨素的得率,采用二次回归正交旋转组合设计对羊软骨硫酸软骨素碱-酶法提取工艺进行了优化,研究了料液比、碱提时间、碱提温度及碱液浓度对硫酸软骨素得率的影响。结果表明,各因素对硫酸软骨素得率影响的顺序为碱提温度>碱液浓度>碱提时间>料液比。在料液比1:6.5、碱提时间2h、碱提温度35℃、碱液浓度7%的条件下,硫酸软骨素得率最高可达12.43%,验证值为12.65%,与理论值基本一致。
In order to enhance the extraction yield of chondroitin sulfate from sheep cartilage,quadratic regression orthogonal design was used to investigated the influence of factors on the yields of chondroitin sulfate such as solvent and solid ratio,extraction time,extraction temperature and alkaline concentration.The results showed that the influencing extent of factors on the extraction yield was extraction temperature,alkaline concentration,extraction time and solvent and solid ratio.When solvent and solid ratio,extraction time,extraction temperature and alkaline concentration were 1:6.5,2 h,35 ℃ and 7 % resepectively,the yield of chondroitin sulfate was 12.43%.The experiment extraction yield was consistent with the theoretical value of 12.65 %.
