高脂饮食诱发大鼠胰岛素抵抗后肿瘤坏死因子-α的改变 被引量:29

摘要 目的 了解高脂饮食所致的大鼠胰岛素抵抗(IR)模型血及组织中肿瘤坏死因子-α(TNF-α)含量的变化。方法 用脂肪占摄入热卡61% 的饮食饲养大鼠7周,以葡萄糖-胰岛素耐量试验(G-InsTT)筛选出IR大鼠,以放射免疫法分别检测其血清、肝脏、红色股四头肌、脂肪组织匀浆中TNF-α的含量。结果 高脂饮食致IR大鼠血及组织中TNF-α含量均明显升高,且其升高程度与高胰岛素血症呈正相关,与G-InsTT中的K值呈负相关。结论 高脂饮食的IR大鼠中存在TNF-α合成和/或分泌的升高。 Objective To study the changes of serum and tissue TNF α concentrations in high fat fed induced rat insulin resistance model.Methods S D rats were fed equicalorically with a high fat diet (fat 61% of total kcal) for 7 weeks, then insulin sensitivity was measured with glucose insulin tolerance test. In rats with insulin resistance, the TNF α concentrations in serum, liver, red quadriceps and adipose tissues were estimated by radioimmunoassay.Results The serum and tissue TNF α concentrations were all significantly increased in high fat fed rats with insulin resistance, and it is positively correlated with serum insulin level and negatively correlated with K values obtained from glucose insulin tolerance test. Conclusion The synthesis and/or secretion of TNF α is increased in high fat fed rats, and is closely correlated with the occurrence of insulin resistance.
