摘要
[目的]研究水介质驯化技术对水稻组培苗的影响。[方法]取500 ml塑料烧杯,加入1/4倍Haogland营养液(内加0.5 mg/LNAA),在烧杯口盖带孔圆盖;取组培苗3~5株,用海绵将其茎基部轻轻裹住,插入烧杯圆盖孔中固定,放到光照培养箱中水介质驯化培养,调整温度、湿度、光照;当组培苗再次有新的根系和根毛时,统计组培苗的成活率,并进行部分生理、分子试验。[结果]水介质驯化水稻组培苗的成活率高,叶绿素含量不低于正常苗的1/4,DNA含量与正常苗接近。[结论]水介质驯化水稻组培苗方法简单易行、节约劳动力、驯化周期短、成活率高,并且可进行相关生理生化、分子试验的测定。
[Objective] The aim was to research water medium domestication technology in rice seedlings of tissue culture.[Method] Firstly,take some plastic beakers for 500 ml and add 1/4 times nutrient solution of Haogland(NAA 0.5 mg/L),then lid a cover with holes on the beakers.Secondly,take 3-5 seedlings from tissue culture and wrapped its stem base gently with sponge,then insert them into the holes of covers.Put the seedlings into the illuminating incubator to cultivate by adjusting temperature,humidity and light.[Result] Domestication seedlings of water medium had high survival rate.Chlorophyll content was not less than 1/4 of chlorophyll content of normal seedlings.DNA content was similar.[Conclusion] Water medium domestication was simple and labor-saving.Domestication cycle was short.Water medium domestication had high survival rate.Domestication seedlings could be used in the related physiology,biochemistry and molecular experiments.
