摘要
本文通过裂褶菌发酵培养获得β-1,3-葡聚糖酶,并对其发酵条件进行了研究。分别探讨了培养基中的碳源、氮源及无机盐对产酶的影响,并采用正交试验找到最佳的产酶培养基配方为葡萄糖2%,牛肉膏0.3%,NH4Cl0.1%,KH2PO40.05%,MgSO40.05%;对培养温度、起始pH值、接种量和培养时间等条件进行了优化,得到该菌株产β-1,3-葡聚糖酶的最佳培养条件为:初始pH值为7.0,温度为30℃,接种量为100mL培养基中(250mL三解瓶)接入2.0mL(孢子浓度为106个/mL),培养时间为6 d。在最佳发酵条件下β-1,3-葡聚糖酶活为15.09U/mL,内切β-1,3-葡聚糖酶活为4.35U/mL。
β-1-1, 3-Glucanase was produced by fermentation of Schizophyllum commune Fr. and the fermentation conditions were optimized. The effect of substrate composition, such as carbon, nitrogen sources, inorganic salts, on the enzyme production were investigated, the optimum substrate was glucose 2%, beef cream 0.3%, NH4Cl 0.1%, MgSO40.05%, KH2PO40.05%. The optimum fermentation conditions was as follows: initial pH 7.0, inoculum concentration 2 mL to 100 mL medium volume in 250 mL triangle flask, temperature 30 ℃, the fermentation period was 6 days. The results indicated that the activity of β-1,3-glucanase could reach 15.09 U/mL, and the activity of endo-β-1,3-glucanase could reach 4.35 U/mL when the Schizophyllum commune Fr was cultured in optimum fermentation conditions.
