肝癌干细胞中差异表达miRNAs对肝癌干细胞增殖和转移的影响 被引量:4

摘要 目的:探讨肝癌干细胞较卵园细胞中高表达的miRNAs(hsa-miR-199a-3p,hsa-miR-122)对肝癌干细胞增殖和转移的影响。方法:分离培养肝癌干细胞,构建并转染肝癌干细胞中差异表达的miRNAs寡核苷酸,通过MTT实验、裸鼠成瘤实验、裸鼠脾脏转移实验观察干扰hsa-miR-199a-3p,hsa-miR-122的表达后肝癌干细胞增殖和转移能力。结果:采用寡核苷酸干扰hsa-miR-199a-3p,hsa-miR-122的表达后,肝癌干细胞增殖能力较对照组显著减弱,裸鼠成瘤实验中形成的肿瘤显著小于对照组,脾脏肿块及肝转移肿瘤显著小于对照组。结论:hsa-miR-199a-3p,hsa-miR-122能够增强肝癌干细胞增殖,促进其成瘤及转移。 Objective To investigate the The effect of differentially expressing miRNA-214 of liver cancer stem cell cycle on cell proliferation and metastasis.Methods Oligonucleotides of hsa-miR-199a-3p,hsa-miR-122 were transfected into cultured liver cancer stem cell.MTT assay,nude mice tumor build up experiment,spleen metastasis assay were performed to evaluate cell proliferation,ability to build up tumor,and metastasis.Results In contrast to the control group,proliferation,ability to build up tumor,and metastasis of liver cancer cells transfected with miRNAs oligonucleotides were signi?cantly reduced.Conclusion Proliferation,ability to build up tumor,and metastasis of liver cancer cells were signi?cantly promoted by hsa-miR-199a-3p and hsa-miR-122.
