摘要
目的观察缺血预处理大鼠心肌中脑源性神经生长因子(BDNF)的mRNA及蛋白表达情况,并探讨其意义。方法 54只雄性SD大鼠随机分为空白对照组(Con组)、缺血再灌注组(I/R组)和缺血预处理组(IPC组),I/R组及IPC组模型建立后,再分别分为再灌注后1、3、6h及12h各时间点亚组。RT-PCR检测心肌组织BDNF mRNA表达,Western-blot法检测心肌组织BDNF蛋白表达。结果 I/R组及IPC组各时间点大鼠心肌组织中BDNF mRNA及蛋白的表达与Con组相比,差异均有统计学意义(P<0.05);而I/R组与IPC组再灌注后同时间点比较大鼠心肌组织中BDNF mRNA及蛋白表达的差异也均有统计学意义(P<0.05)。结论缺血预处理可以上调BDNF的mRNA及蛋白表达,推测BDNF在缺血预处理后对缺血再灌注大鼠心肌具有保护作用。
Objective To observe BDNF mRNA and protein expression in the myocardial ischemic precondi tioning and its significance. Methods 54 male SD rats were randomly divided into control group (Con group), ischemiareperfusion group ( L/R group) and ischemic preconditioning group ( IPC group). After establishment of I/R group and IPC group model, all rats weredivided into lh, 3h, 6h and 12h group after reperfusion. BDNF mRNA were detected by RT-PCR and the BDNF protein were detected by Western-blo. Results Compared with the Con group, the expression of BDNF mRNA and protein in I/R group and IPC group at each time in rats showed significant differences ( P 〈 O. 05 ) ; and compared I/R group with IPC group, the expression of BDNF mRNA and protein at same time in rats showed significant differences (P 〈 0. 05). Conclusion Ischemic preconditioning can increase the expression of BDNF mRNA and protein. BDNF after the ischemic preconditioning has a protective effect on myocardial ischemia -reperfusion.
