青蒿琥酯对MDA—MB-231细胞增殖抑制作用及其机制

摘要 目的探讨青蒿琥酯(artesunate,ART)对乳腺癌雌激素受体阴性细胞MDA—MB-231抑制作用及其机制。方法ART处理细胞3d后,采用MTT比色法检测细胞增殖功能,观察细胞形态、结构的改变,免疫细胞化学检测Bax,nm23,Bcl-2,P21WAF1/CIP1蛋白的表达情况。结果ART作用于乳腺癌细胞MDA—MB-231后,发现其对细胞的抑制作用呈明显浓度依赖性,可导致细胞形态、结构改变;免疫细胞化学结果显示2μmol/L青蒿琥酯作用细胞72h后,药物组同细胞对照组比较,可上调Bax、nm23、P21WAF1/CIP1蛋白表达,差异有统计学意义(P〈0.05),Bcl-2蛋白质表达差异无统计学意义(P〉0.05)。结论ART有抑制MDA—MB-231细胞增殖的作用,其机制可能与上调Bax、nm23、P21WAF1/CIP1蛋白表达有关。 Objective To study the effects of artesunate (ART) on estrogen receptor negative breast cancer cell line MDA-MB-231 and its mechanisms. Methods Treated with ART for 3 days, MDA-MB-231 cells proliferation was examined by MTT assay. The morphological and uhrastructural changes of MDA-MB-231 cells were observed under microscope and electronic microscope. Immunocytoehemistry was used to detect the expression of Bax, nm23, Bcl-2, and P21WAF1/CIP1. Results ART treatment led to a dose dependent inhibition of MDA-MB-231 cells. ART could change the morphology and ultrastmcture of MDA-MB-231 cells. After treatment with ART (2μmol/L) for 72 hours, immunocytochemical staining showed that the expression of Bax, nm23, and P21WAF1/CIP1 was upregulated in comparison to the control group (P 〈 0. 05) while the expression of Bcl-2 in MDA-MB-231 cells didn't have a significant change ( P 〉 0. 05 ). Conclusions ART shows an anti-proliferative effect on MDA-MB-231 cells. The mechanisms may be related to upregulation of Bax, nm23 and P21WAF1/CIPI expression.
