摘要
目的初步探讨人乳腺癌细胞MCF-7经Uv-c照射后PTEN表达及分布变化。方法利用流式细胞仪检测Uv-c照射后细胞周期改变;利用Westernblot检测Uv-c照射后PTEN表达变化,并且利用免疫荧光方法检测PTEN细胞分布变化。结果 UV-c照射前,细胞内PTEN主要分布于胞核周围的胞质中;UV-c照射MCF-7细胞后,使阻滞于细胞G2/M期,24后,细胞内总PTEN量明显增多,且主要均匀地分布在细胞质中。结论 Uv-c照射后,细胞处于周期阻滞状态,伴随着PTEN蛋白表达上调且部分蛋白发生浆核转位。
Objectives To study the expression and distribution of PTEN in breast cancer cell MCF-7 after Uv-c irradiation.Methods The cell cycle distribution was examined using flow cytometry.The protein expression was detected with immunoblot analysis.The protein location was detected with immunofluorescence analysis.Results Before UV-c irradiation,the PTEN cells mainly distributed in the cytoplasm surrounding the nucleus;After 24 hours of irradiation,the MCF-7 cells were arrested at G2 / M phase,and total PTEN cells were significantly increased,and evenly distributed mainly in the cytoplasm.Conclusions Uv-c irradiation could cause cell cycle arrest in human MCF-7 cell.In the meantime the levels of of PTEN was increased and some of them shifted into the cell nucleoli.
