摘要
目的:探索合适的小凹制作方法以培养小鼠无透明带胚胎,完善WOW(the well of the well)系统,为手工克隆技术提供支持,提高核移植生产效率.方法:对WOW系统进行改进,用金属针、玻璃针及胚胎聚集针分别做小凹,比较这三种方法制作的小凹对小鼠无透明带受精胚胎的培养效果.结果:胚胎聚集针所做小凹的囊胚发育率显著高于金属针及玻璃针所做小凹(55.61%vs44.38%,43.16%).结论:胚胎聚集针制作的小凹培养小鼠无透明带受精胚胎,囊胚发育率最高.
The aim of the research is to search for suitable methods of making wells, for culturing the mouse zona -free embryos, and to modify the WOW culture method to improve the efficiency of the handmade cloning technique. In order to improve the WOW system, we use the metal needle, the glass needle, and the embryos aggregation needle to make wells respectively, and compared the culture effects of the zona -free embryos in those wells. Blastocyst development rate in the wells made by embryos aggregation needle was significantly higher than that in the metal needle and the glass needle wells(55.61% vs 44.38% ,43.16% ). The zona - free embryos that cultured in the wells made by embryos aggregation needle showed the hightest blastocyst development rate.
