摘要
目的:探讨心理应激源对脑组织Na+ ,K+ - ATP酶活性的影响及发生机制,以及自由基清除剂对该酶活性的保护作用。方法:将大鼠分为对照组、束缚组和治疗组。用酶组化法制片,用图像分析仪测脑组织Na+ ,K+ - ATP酶的活性;用TAB法测MDA的含量。结果:束缚组和治疗组的Na+ ,K+ - ATP酶活性明显低于对照组,MDA含量明显高于对照组;治疗组Na+ ,K+ - ATP酶活性明显高于束缚组,MDA含量明显低于束缚组;束缚组、治疗组的MDA含量与Na+ ,K+ - ATP酶活性呈直线负相关。结论:较长时间的束缚应激可致大鼠脑组织Na+ ,K+ - ATP酶活性降低;该酶活性降低与氧自由基损伤有关;给予SOD+ CAT治疗。
To study the mechanisms of effect on activity of Na +,K +-ATPase in brain tissue by psychic stressors and the protecting function of free radical enzymatic scavengers. Methods:Experimental rats were divided into 3 groups: control group, immobilization group and group of administration of SOD and CAT (administration group). Section preparations were prepared by enzymo-histochemistry. Activity of Na +,K +-ATPase in Brain tissue was indicated by grey scale value detected by an image-analysing instrument. MDA content in brain tissue was measured by TBA method and protein in the tissue was quantitatively analysed by biuret method.Results: Activity of Na +,K +-ATPase is lower while MDA content is higher in immobilization and administration groups than in control group. Activity of Na +,K +-ATPase is obviously higher while MDA content is lower in administration group than in immobilization group. MDA content and activity of Na +,K +-ATPase are rectilinear and negatively correlative. Conclusion: Long-term immobilization stress caused decrease in activity of Na +,K +-ATPase in rat brain tissue. The decrease is relevant to lesion of oxygen free radicals. SOD and CAT had protective function in preserving activity of Na +,K +-ATPase.
