摘要
蛋白质提取是进行蛋白组学分析的关键步骤。本试验比较了水溶、盐溶及醇溶3种提取方法对获取梅花鹿鹿茸总蛋白含量及SDS-PAGE电泳分辨率等的区别。结果表明,水溶法提取鹿茸蛋白质最佳,该方法提取的蛋白质含量高、能得到比较好的SDS-PAGE电泳图谱。进而对SDS-PAGE电泳操作步骤进行了探讨,确立了适用于蛋白组学下游技术-双向电泳分析(蛋白质提取、凝胶制备、染色、脱色)的方法。
Protein extraction from tissue or cells is a key step to achieve proteome analysis.In this study,three protein extraction methods based on water(Millipore water),brine(sodium chloride) and alcohol(isopropyl alcohol),respectively,were compared,in order to acquire total protein from sika deer antlers,furthermore,to establish a clear SDS-PAGE map of sika deer antlers.It was found that brine was the best one resulting in the highest total protein yield,the most clear in SDS-PAGE.Moreover,the further discussion was made to the SDS-PAGE electrophoresis sequence of operation.The successful establishment of the extract technique lays the foundation for the analy-sis methods in proteomics study downstream technology—two-dimensional electrophoresis.
