黄芪皂苷Ⅳ对大鼠脑缺血/再灌注后海马血管生成的影响 被引量:9

The Effect of Astragaloside Ⅳ on Angiogenesis in Hippocampal Region after Cerebral Ischemia-reperfusion in Rats

摘要 目的探讨黄芪皂苷Ⅳ对大鼠缺血/再灌注损伤后海马中血管内皮生长因子(VEGF)和微血管密度(MVD)表达的影响。方法实验动物随机分为假手术组、模型组、尼莫地平对照组、黄芪皂苷Ⅳ组。缺血2h再灌注72h后,各组进行神经功能评分,采用Westernblot及免疫组化方法检测VEGF蛋白表达,免疫组化法进行CD34染色计数MVD。结果黄芪皂苷Ⅳ组神经功能损害评分低于模型组和尼莫地平对照组(P<0.05);与尼莫地平对照组和模型组比较,黄芪皂苷Ⅳ组可明显上调VEGF蛋白表达及MVD(P<0.05)。结论黄芪皂苷Ⅳ对大鼠脑缺血/再灌注有神经保护作用,其作用机制可能与促进海马VEGF蛋白表达,增加MVD有关。 Objective To investigate the effect of astragaloside Ⅳ on vascular endothelial growth factor(VEGF) and microvessel density(MVD) in hippocampal region after cerebral ischemia-reperfusion(CIR) in rats.Methods Rats were randomly divided into sham operation group,model group,nimodipine group and Astragaloside Ⅳ group.72 hours after reperfusion,neurological function score were observed,MVD was evaluated by immunohistochemistry with the specific antibody CD34.The expressions of VEGF proteins was observed by using immunohistochemical and Western blot.Results Compared with model and nimodipine group,the neurological scores of astragaloside Ⅳ group was significantly decreased.Compared with model and nimodipine group.The expression of VEGF protein and MVD were significantly increased in astragaloside Ⅳ group.Conclusion Astragaloside Ⅳ could improve the neurological function in rats after cerebral ischemia-reperfusion,which may be related to up-regulating VEGF protein and MVD in hippocampal region.
